Immune fibroblastic cell (IFC) subpopulations of dermal fibroblasts differentiate into adipocytes and provide skin immune defense in response to infection. To better understand the role these fibroblast subsets, we have undertaken a systematic analysis models inflammation. First, communication from mesenchymal, epithelial, bone-marrow derived cells was investigated using unbiased machine learning single-cell RNA sequencing (scRNAseq) data mice infected with Cutibacterium acnes. This network revealed that communicate more myeloid than any other lineage. Further scRNAseq showed lipocalin 2 (LCN2/NGAL)—a chemoattractant antimicrobial protein—was major gene upregulated by IFCs (17-fold change, p<0.0001), LCN2/NGAL most highly expressed an IFC subset genes related fat differentiation.
